Přednáška seznamuje posluchače s biologií kvasinek v plném rozsahu. Zahrnuta jsou témata: cytologie, buněčný cyklus, stárnutí, apoptóza, konjugace, sporulace, killer fenomén, jaderný a mitochondriální genom, priony, transposony, metabolismus, taxonomie, patogenní kvasinky, kvasinky v průmyslu a v přírodě. Důraz je kladen na nejnovější poznatky kvasinkové genetiky a molekulární biologie. Velmi doporučuji zapsat si současně i nepovinný praktický kurz (Praktikum z molekulární a buněčné biologie kvasinek - MB140C29).
Doporučuji předchozí absolvování přednášky Základy molekulární biologie a Mikrobiologie.

The lecture covers the wide area of the yeast biology: yeast cytology, regulation of the cell cycle,aging, apoptosis, mating, meiosis and sporulation, killer phenomenon, nuclear and mitochondrial genome, prions, transposons, yeast metabolism , taxonomy, yeast as pathogens, yeasts in industry, ecology of yeasts. The emphasis is laid on recent progress in understanding of the yeast genetics and molecular biology.
It is recommended to enroll on a practical course (Practical course in molecular and cell biology of the yeasts - MB140C29).
It is recommend previous completion of the lecture Essentials in molecular biology and Microbiology.

Exam requirements - knowledge of the content of lectures. The exam is written, containing 3 questions where a more extensive answer is expected - each question is rated max. 8 points and 10 questions where only a very brief answer is required - here a maximum of 2 points can be obtained for each answer. It's 1 hour to work out the answers. If interested, an oral exam can be arranged.

1. Úvod.
Definice kvasinek, význam, využití. Požadavky na prostředí - voda, teplo, kyslík. Kultury kvasinek v tekutém a na pevném médiu. Izolace kvasinek, eliminace kvasinek. Morfologie kvasinek, typy rozmnožování, typy buněk.
Saccharomyces cerevisiae, Schizosaccharomyces pombe. Fáze buněčného cyklu, start - výběr vývojových alternativ, genová regulace, cykliny, signální dráhy - Ras-cAMP dráha, , polarizovaný růst pupene - výběr místa založení pupene, polarizace růstu, pseudohyfální růst.
6. Stárnutí a apoptóza.
7. Pohlavní rozmnožování kvasinek.
S. cerevisiae, Sch. pombe a další kvasinky. Životní cyklus u basidiomycetových kvasinek. Genetická determinace párovacího typu, homothalismus a heterothalismus, průběh konjugace, feromony, receptory, aglutininy, feromonová signální dráha. Sporulace - morfologické změny, biochemické změny, genetická determinace, spory.
8. Jaderný genom.
Metody studia, sekvenování genomu, variabilita, , transposony. 2 µm plasmidy.
9. Mitochondriální genom.
Velikost a uspořádání mt DNA, genetická mapa, mitochondriální mutanty, respiračně deficitní mutanty, přenos mitochondriálního genomu.
10. Kvasinkové priony.
11. Killer fenomén.
Killer kmeny s dsRNA, cytoplasmatickou DNA, jadernou determinantou, charakteristiky systémů, mechanismy účinku, využití killer fenoménu, pradátorské kvasinky.
12. Metabolismus.
Transport látek do buňky, zdroje uhlíku a energie, zdroje dusíku, zásobní látky. Hlavní regulační dráhy. Metabolické regulace - katabolická represe, Pasteurův, Kluyverův a Custersův efekt. Kvasinkové ATPázy.

Yeast cultures in liquid and on solid medium. Cell morphology, modes of propagation, cell types. 2. Cytology. Capsule, cell wall, scars, flocculation, plasma membrane, secretion pathways, nucleus, vacuole, mitochondria, peroxisomes, cytoskeleton. 3. Cell cycle. Saccharomyces cerevisiae, Schizosaccharomyces pombe. Phases of the cell cycle, Start - selection of developmental possibilities, gene regulation, cyclins, signal pathways, polarised growth of the bud, pseudohyphal growth. 4. Senescence and apoptosis. 5. Sexual reproduction of the yeasts. S. cerevisiae, Sch. pombe, other yeasts. Life cycle of basidiomycetous yeasts. Genetic determination of mating type, homothallism, heterothallism, conjugation, pheromones, receptors, agglutinins, pheromone signal pathway. Sporulation - morphological, biochemical processes, genetic determination, spores. 6. Nuclear genome.
