Laboratory course in gas chromatography is intended especially for students of master and doctoral programmes. It includes five laboratory exercises that will enable students to acquire a deeper understanding of key aspects of gas chromatography, preparation and measurement methods samples, the evaluation of results, including qualitative and quantitative analysis, and the validation of analytical methods.
Literature

McNair H. et al.: Basic Gas Chromatography. 3rd ed. Wiley 2019.
Gas Chromatography. 2nd ed. C. Poole (ed.) Elsevier 2021.
Additional specialized literature will be handed out during laboratory exercises.

Requirements to the exam
Last update: doc. RNDr. Karel Nesměrák, Ph.D. (22.11.2023)

Pass all practicum tasks and the final written test (minimum pass rate 65%).

Syllabus
Last update: doc. RNDr. Karel Nesměrák, Ph.D. (22.11.2023)

1. Introduction to GC. Description of GC main parts, column, liner and septum
installation and exchange procedures. Servicing and cleaning of some GC parts. GC
method creation and optimisation (flow rates, oven temperature, split etc.).
2. External standard method. Calibration and quantitative determination of analyte in the
testing solution. Calculation of basic metrological parameters: SD, RSD, uncertainty
and accuracy.
3. Gravimetrical preparation of standard solutions of volatile compounds. Tips and
advices. Dilution of the prepared solutions, creation of the validation experiments plan
and performing the validation itself. Linearity, precision, accuracy, uncertainty,
sensitivity and other validation parameters evaluation.
4. Continuation of previous work (3). Validation of two Internal Standard methods for the
analysis of alcoholic products and their comparison. Evaluation of the results.
5. SPME method for GC-MS analysis of any selected object. Sample preparation,
extraction, and measurement and GC-MS spectra evaluation. Literature search for
additional processing of the observed results.

Entry requirements
Last update: doc. RNDr. Karel Nesměrák, Ph.D. (22.11.2023)

There are no entry requirements, however, it is expected that student have completed a course in analytical chemistry and a lecture in separation methods.
