1 / Preparation of culture media and sterilization. Types and composition of culture media, sterilization methods, preparation of sterile glass and instruments, disposal of used experimental material. 2 / Vaccination and aseptic work. Aseptic techniques, work with pure culture, isolation of microorganisms, enrichment techniques, cultivation conditions. 3 / Native and permanent preparations, Gram staining of bacteria, staining of spores. 4 / Quantitative tasks with microorganisms. Determination of number of live bacteria in acidophilic milk by sowing on solid culture medium, monitoring of presence of coliform bacteria in water samples. Determination of the total number of microorganisms, counting chamber. 5 / Approaches to identification of bacteria. Monitoring of biochemically important properties of bacteria, identification of unknown enterobacteria by commercial test (Enterotest). 6 / Influence of environmental factors (pH) on the growth of microorganisms, determination of optimal pH for selected strains of bacteria and yeasts.
