Structure and function of the plant cytoskeleton. Methods for cytoskeleton studies. Tubulin and actin - biochemical properties, isoforms and posttranslational modifications. Associated proteins - characterization, localization, function. Changes of cytoskeleton in plant cell cycle. Motor proteins, vesicle and organelle transport. Gene for plant cytoskeleton - localization, expression and regulation. Construction, selection and utilization of genotypes with modiefied cytoskeleton. The role of cytoskeleton in the plant morphogenesis including ontogenetic and organ specifity.

Metody studia cytoskeletu

 

Aktin a mikrotubuly: základní stavba, monomery, vlastnosti monomerů a polymerů, tvorba vláken a depolymerace, studium in vitro a in vivo. Izotypy.

Asociované proteiny s aktinem

 

Asociované proteiny s mikrotubuly

 

Role aktinu: cytoplazmatické proudění, pohyb. Endocytóza. Buněčná stěna, polární růst. Gravipercepce.

 

Role mikrotubulů: buněčný cyklus, syntéza BS, bičíky. MTOC.

 

Dělení buňky: mitóza, meióza, fragmoplast.

 

Bičíky a řasinky, centrozóm a BB, blefaroplast.

 

Morfogeneze a cytoskelet: známé procesy tvarování trichomů, pokožkových buněk, vrcholový růst. Endocytóza a pohyb molekul v rámci endozómu?
