Practical Course of DNA diagnostics Annotation: The aim of this practical course is to introduce recent basic techniques used in DNA diagnostics. After theoretical opening all students will have the possibility to try practically basic methods in lab. Course is designed for highly motivated students of 3rd to 6th year of general medicine studies, who are thinking about PhD studies; and is open also for PhD. students. Course is open for 6 students; takes 5-6 hours of two afternoons, and takes place in Laboratoř pro studium mitochondrialnich poruch, at the Dep. of Pediatrics (Klinika dětského a dorostového lékařství), Ke Karlovu 2, Praha 2, building E4. Notice: The exact date is arranged individually due to the signed students’ possibilities. Syllabus:

Practical course of DNA diagnostics.

Scope of course: theoretical opening; DNA isolation (classical methods and commercial kits) from different tissues (blood, hair, buccal cells, cultivated fibroblasts), concentration measurement (NanoDrop), methods of DNA amplification (PCR- practically; cloning - theoretically), mutation detection methods - implications and applications (practically: ELFO, RFLP; theoretically: sequencing, HA, DGGE). Molecular results interpretation.
