Designed for those interested in histological methodologies, suitable as a supplement lectures on histology. Recommended to take the course first year of bc. study after finishing the Practicals of Cell Biology. This is a basic course of histological techniques, where students learn about preparing smearing, stemping and cutting types of histological specimens, including fluorescent variant of staining.

Určeno pro zájemce o histologické metodiky, vhodné jako doplněk přednášky z Histologie. Doporučený ročník 1. po absolutoriu praktik z Buněčné biologie. Jedná se o základní kurs histologických technik, kde se studenti seznámí s přípravou roztěrových, otlakových a řezových histologických preparátů, včetně fluorescenčních variant.

For this practical is not required any special literature, protocols are provided. At the beginning of the practical exercises all issues are thoroughly discussed and all protocoarer are available.

Pro absolutorium tohoto praktického cvičení není vyžadována žádná literatura, protokoly jsou součástí pracovních stolů, na začátku praktického cvičení jsou pečlivě probrány všechny protokoly, všetně teorie histologických metod, nezbytné pro úspěšné zvládnutí praktických cvičení.

For getting the credits the 100% attendance is obligatory. Students have to prepare set of required specimens and their images using a microscope equipped with a color camera with high resolution - both in the fluorescence mode and in transmitted light.

Pro udělení zápočtu je nezbytná stoprocentní docházka, příprava požadovaných preparátů a jejich zachycení pomocí mikroskopu vybaveného barevnou kamerou s vysokým rozlišením - a to jak ve fluorescenčním modu, tak v procházejícím světle.

1. Preparation of histological specimens: the procurement of tissues and organs (mouse), fixation, drainage using ascending series of alcohol, embedding in paraffin, cutting on a microtome, staining with hematoxylin / eosin, closing in Canadian balsam. 2. Preparation of smearing specimens of blood and bone marrow: methanol fixation, Giemsa 3. Preparation of stamping specimens of selected organs - spleen, thymus, lymph nodes, testes, cerebellum fixation with methanol, Giemsa 4. Preparation of fluorescently labeled specimens (detection of plasma cells in the spleen, characterization of the developmental stages of spermatogenesis in the testis) 5. Preparation of immunofluorescent slides for fluorescence microscopy (fixation, permeabilization blocking, staining with primary and secondary antibodies, DAPI, embeding in Mowiol)
