Annotation -
The laboratory course on separation methods is focused on gaining theoretical knowledge as well as experimental skills in advanced analytical instrumentation. Students will solve four complex problems using high-performance liquid chromatography with tandem mass detection, gas chromatography with mass or flame ionization detection (student's choice), capillary zone electrophoresis, and chiral capillary electrophoresis.

After passing this course students will be able in capillary electrophoresis, liquid chromatography with tandem mass spectrometry, and gas chromatography with flame-ionization or mass detection:
Independently operate instruments and perform analyses on the basic user level with written instructions.
Evaluate and interpret the experimental records.
Draw appropriate conclusions from the results.
Create a concise report containing a description of the experimental work, results, and conclusions, without serious factual or formal errors.

Participation in the practical exercises, submission of appropriate reports, examinations for individual tasks, and scoring minimum 60% in the final test.

Syllabus -

1) Three amino acids will be separated using a cyclodextrin-based chiral selector by capillary electrophoresis. The migration order of individual enantiomers will be determined. Enantiomers will be quantified using the standard addition method.
2) An HPLC-MS-MS method for analysis of selected organic pollutants will be developed. Focus will be on the optimization of MS/MS detection.
3) This exercise comprises gas chromatography with flame ionization or mass spectrometry detection. Students can choose according to their interests.
4) Ammonium, sodium and potassium ions in urine are separated and determined using capillary electrophoresis.
