Přednáška podává přehled o struktuře a funkci cytoskeletu rostlinné buňky. Informuje o metodách studia cytoskeletálních proteinů, o biochemických vlastnostech mikrotubulů, mikrofilament, intermediálních filament a asociovaných proteinů. Zabývá se účastí cytoskeletu v morfogenezi rostlin, zejména rolí v buněčném cyklu, syntéze buněčné stěny a transportu. Zahrnuje i reakce CS na různé typy signálů, vliv xenobiotik (zejména těžkých kovů) a faktorů vnějšího prostředí. Představuje i nejnovější poznatky o tubulinových genech, cytoskeletálních mutantách a drogách, včetně jejich využití.

Characterization of the cytoskeletal components. Basic function of the cytoskeleton. Nuclear and membrane skeleton. Evolution of the cytoskeleton.
2. The basic metodical approaches to the cytoskeleton studies. I.
Immunocytological and immunohistochemical approaches. Electron and confocal microscopy. GFP techniques.
3. The basic metodical approaches to the cytoskeleton studies II
Izolation and characterization of the cytoskeletal proteins. Microinjection techniques. Simulation experiments in vitro. Cytoskeletal toxins. Genetical methods and cytoskeleton mutants.

Účast cytoskeletu ve vnitrobuněčném transportu, proudění cytoplasmy a transportu organel. Molekulární motory. Role aktinu v mezibuněčném transportu. Plasmodesmy.
8. Funkce cytoskeletu III.
Geny pro cytoskelet - lokalizace, stabilita, exprese a regulační faktory. Konstrukce, selekce a využití genotypů s modifikovanou stabilitou cytoskeletu. Cytoskelet v morfogenezi rostlin, ontogenetická a orgánová specifita. Odpověď na signály - světlo, mechanické a gravitační stimuly, fytohormony.
9. Funkce cytoskeletu IV.
Účast cytoskeletu při tvorbě buněčné stěny. Interakce mikrotubulů, mikrofilament a plasmatické membrány. Deposice celulózových mikrofibril zprostředkovaná mikrotubuly.Transport Golgiho vesikulů.
10. Funkce cytoskeletu V.
Cytoskelet při růstu a vývoji rostlin - polarita, elongace a expanse buněk, tvorba listových primordií, průduchů, trichomů, kořenových vlásků, hlíz a cibulí, stonků, pylových zrn a pylových láček, změny cytoskeletu při stárnutí a apoptóze.
11. Funkce cytoskeletu VI.
Vliv xenobiotik a stresových faktorů na strukturu a funkci cytoskeletu. Příklad odpovědi cytoskeletu na abiotický a biotický stres - extrémní teploty, těžké kovy, napadení houbovými a virovými patogeny.
