Advanced molecular techniques are now routinely used in the microbiological laboratory. Student of this course will thus learn common molecular methods, such as PCR and its modifications, MALDI-TOF, next generation sequencing (NGS) and others utilized in microbiology. In addition, the students will acquire knowledge about specific incorporation of these methods in the clinical diagnostic procedures. During the first practical, the students will perform an isolation of nucleic acids, PCR setup and subsequent detection using gel electrophoresis. The second practical will aim at performing an identification of bacterial cultures using MALDI-TOF.

Seminars

 1.    Polymerase chain reaction (PCR) and its modifications (3hrs) – theory of PCR, both classical and advanced modifications

2.    Sequencing (3hrs) - theory of sequencing reactions, Sanger sequencing, NGS and their use in the microbiological diagnosis and research

3.    MALDI-TOF and its applications (3hrs) - principles and methodology of mass spectrometry

 

Practical classes

 1.    PCR - isolation of nucleic acids, setting up a PCR reaction, detection of products using gel electrophoresis (3-4hrs)

2.    MALDI-TOF - practically performing an identification of cultures (3hrs)
