Students who have passed histology, embryology and biology are eligible to register. In lectures, students gain knowledge in a progressively developing field of stem cell biology and regenerative medicine incl. closely related areas of tissue engineering and cell therapy. In the course of practical classes, students become familiar with detection methods and basic procedures in the tissue culture laboratory.

1    Introduction. Regenerative medicine, regeneration, reparation. Cell therapy. Stem and progenitor cells - basic terms, characterization, biological behaviour, differentiation potential. Stem cell plasticity. Telomeres, telomerase and ageing of stem cells. Ethics and moral aspects of stem cell research. (Mokry, 3)
2    Pluripotent stem cells, ES cells, iPS cells. Epithelial stem cells. Tumour stem cells. Muscle-derived stem cells; muscle regeneration. (Mokry, 3)

3    Neural stem cells and their niche. Regeneration of the nervous tissue. Spinal cord injury. (Mokry, 3) 
4    Endothelial progenitor cells. Haematopoietic stem cells. Haematological applications, bone marrow transplantation. (Mokry, 1; Filip, 1; Žák, 1)

Practical courses
1 Tissue engineering. Laboratory facilities and basic procedures in tissue culture laboratory. Safety aspects, risk assessment. Preparation of buffers and media. Phase contrast microscopy.
2 Cultivation techniques. Isolation of cells from human and animal tissues. Enzymatic tissue dissociation. Cell quantification. Cryoconservation. Mathematics in the laboratory - number of population doublings, doubling time, kinetics.
3 Cell isolation. Primary cultures. Subculturing. Cultivation surfaces. Basic biological characteristics. Cell separation techniques. Gradient centrifugation (Ficoll, Percoll). Immunomagnetic separation.
4 Cultivation protocols. Stem cell differentiation in vitro. Extracellular matrix analysis.
5 Visit of tissue culture laboratory. Cell viability. Preparation of cells for flow cytometric analysis.
