Preliminary scope of work
Acinetobacter baumannii je významný nemocnicní patogen, který zahrnuje kmeny
rezistentní k vetšine nebo i všem terapeuticky využitelným antibiotikum. Tato rezistence je
výsledkem rady bunecných mechanizmu, mezi nimiž hrají zásadní roli systémy aktivního
efluxu antimikrobních látek. Tato práce podává prehled jednotlivých rodin efluxových
systému a jejich dosud známých zástupcu u rodu Acinetobacter. Zameruje se na efluxový
systém AdeABC náležející do rodiny RND (resistance-nodulation-cell division family). Tento
systém je prítomen u vetšiny kmenu A. baumannii a tvorí jej tri strukturní proteiny, jež jsou
regulovány dvousložkovým regulacním systémem. Nadprodukce AdeABC zpusobuje
rezistenci nebo snížení citlivosti k rade antibiotik a je obvykle spojená s dalšími mechanizmy
rezistence.
Preliminary scope of work in English
Acinetobacter baumannii is an important nosocomial pathogen which includes also
strains resistant to majority or all therapeutically relevant drugs. This resistance is a result of
the activities of many cellular mechanisms, among which systems of active efflux of
antimicrobial agents play an important role. This study describes all main families of bacterial
efflux systems and their known representatives described in the genus Acinetobacter. It
focuses on the efflux system AdeABC belonging to the RND family (resistance-nodulationcell
division family). This system is present in the majority of A. baumannii strains and it is
formed by three structural proteins regulated by a two-component system. Overexpression of
AdeABC results in resistance or decreased susceptibility to many drugs and is usually
associated with other resistance mechanisms.
