Cílem práce bude vývoj nové chemické metody, která umožní kovalentní značení proteinů specifickými ligandy a jejich následné uvolnění. Pro tuto funkci budou zváženy a testovány různé chemické návrhy. Připojení tohoto modulu "chyť a pusť" do knihovny ligandů umožní efektivní objevování specifických ligandů pro různé proteiny a fenotypy. První část práce bude sestávat z organické syntézy modulů "chyť a pusť" a testování jejich funkčnosti. Druhá část bude zaměřena na aplikaci konceptu ve vysoce výkonném objevu proteinových ligandů. Školitel je připraven studentovi zajistit minimální měsíční příjem požadovaný novelou vysokoškolského zákona.
Preliminary scope of work in English
The aim of the work will be the development of a new chemistry that will enable covalent labelling of proteins with specific ligands and their subsequent release. Various chemical designs for this functionality will be considered and tested. Attachment of this catch-and-release module to a ligand library will enable efficient discovery of specific ligands for various proteins and phenotypes. The first part of the work will consist of organic synthesis of the catch-and-release modules and testing their functionality. The second part will be focused on the application of the concept in high throughput discovery of protein ligands. The supervisor is prepared to provide the student with the minimum monthly income required by the amendment to the Higher Education Act.
