Torque teno virus (TTV) patří do skupiny Alphatorquetenovirus z čeledi Anelloviridae. Tato čeleď zahrnuje velmi heterogenní skupinu malých neobalených DNA virů s jednořetězcovým kruhovým genomem o velikosti od 2,8kb do 3,8kb. TTV představuje nejrozšířenější složku lidského viromu. Hlavním místem replikace viru jsou pravděpodobně lymfocyty, i když virová DNA byla detekována i v jiných typech buněk. Virus je přítomen v krvi zdravých jedinců v relativně nízké virové náloži díky kontrole replikace viru imunitním systémem hostitele. U imunosuprimovaných pacientů dochází vlivem nedostatečné funkce imunitního systému k výrazným změnám virové nálože. Z tohoto důvodu může TTV sloužit jako biomarker stavu imunitního systému. Cílem této bakalářské práce bude popsat TTV a zhodnotit jeho roli imunitního markeru nejenom u pacientů po transplantaci, ale i jedinců s autoimunitním onemocněním a u HIV pozitivních osob. |

Torque teno virus (TTV) belongs to the Alphatorquetenovirus group of the Anelloviridae family. This family comprises a very heterogeneous group of small uncoated DNA viruses with single-stranded circular genomes ranging in size from 2.8kb to 3.8kb. TTV represents the most abundant component of the human virome. The main site of viral replication is probably lymphocytes, although viral DNA has been detected in other cell types. The virus is present in the blood of healthy individuals at a relatively low viral load due to the control of viral replication by the host immune system. In immunosuppressed patients, significant changes in viral load occur due to inadequate immune function. For this reason, TTV may serve as a biomarker of immune status. The aim of this bachelor thesis will be to describe TTV and evaluate its role as an immune marker not only in post-transplant patients, but also in individuals with autoimmune disease and in HIV positive individuals. |
