Preliminary scope of work

Schizofrenie je vývojové neuropsychiatrické onemocnění s komplexní symptomatikou. Vedle pozitivních a negativních symptomů je zásadním projevem i kognitivní poškození. Právě narušení kognitivních funkcí a remediace tohoto narušení je možné studovat u animálního modelu schizofrenie.
Cílem navržené diplomové práce je studovat kognitivní narušení v animálním modelu schizofrenie (po aplikaci non-kompetitivních agonistů glutamátu), případně při souběžné tryptofanové depleci, která modeluje afektivní poruchy. Abychom minimalizovali interindividuální rozdíly v chování, pro animální model budou použiti dospělí samci potkana (Rattus norvegicus).
Zaměříme se na sociální chování, paměť a kognitivní flexibilitu a jejich změny v modelu schizofrenie. Důležitým bodem bude také zavedení úlohy časové percepce v intervalovém časování a studium jeho změn v modelu schizofrenie. Právě narušená časová percepce u pacientů trpících schizofrenií může představovat podklad pro komplexnější kognitivní změny, dezorganizovanost a fragmentované myšlení. Pochopení kognitivního narušení u schizofrenie umožní lépe pochopit mechanismy onemocnění i identifikovat případné terapeutické cíle.

Preliminary scope of work in English

Schizophrenia is a developmental neuropsychiatric disorder with complex symptomatology. In addition to positive and negative symptoms, cognitive impairment is a significant manifestation. It is the cognitive impairment and the remediation of this impairment that can be studied in the animal model of schizophrenia.
The aim of the proposed thesis is to study cognitive impairment in the animal model of schizophrenia (after the administration of non-competitive glutamate agonists), possibly with simultaneous tryptophan depletion, which models affective disorders. To minimize interindividual differences in behaviour adult male rats (Rattus norvegicus) will be used for the animal model of schizophrenia.
We will focus on social behavior, memory and cognitive flexibility and their changes in a model of schizophrenia. Importantly, we will also introduce the task for testing time perception in interval timing and study its changes in a model of schizophrenia. It is the impaired time perception in schizophrenia patients that may underlie more complex cognitive changes, disorganization and fragmented thinking. Understanding cognitive impairment in schizophrenia will allow a better understanding of the mechanisms of the disease and the identification of potential therapeutic targets.
