Preliminary scope of work
Náplní práce bude příprava nabohacené cytoskeletální frakce zástupců skupiny Preaxostyla - Monocercomonoides exilis a Paratrimastix pyriformis - a následná charakterizace jejího proteinového složení pomocí hmotnostní spectrometrie. Cílem je odhalit proteinové komponenty některých z cytoskeletálních struktur asociovaných s mikrotubulárními kořeny, zejména I fibrily (preaxostyl), B-, C-fibrily a dalších. Proti nadějným kandidátům budou vytvořeny protilákty s cílem ověřit jejich lokalizaci pomocí imunofluorescence.
Preliminary scope of work in English
The scope of work will be the preparation of an enriched cytoskeletal fraction of the Preaxostyla flagellates - Monocercomonoides exilis and Paratrimastix pyriformis - and subsequent characterization of its protein composition by mass spectrometry. The aim is to reveal the protein components of some of the cytoskeletal structures associated with microtubular roots, especially I fibrils (preaxostyl), B-, C-fibrils and others. Antibodies will be generated against promising candidates in order to verify their localization by immunofluorescence.
