Karcinom z Merkelových buněk (MCC) je vzácný agresivní kožní neuroendokrinní nádor s častým výskytem metastáz. Více než 80% MCC je asociováno s infekcí lidského polyomaviru Merkelových buněk (MCPyV). Zásadním krokem v karcinogenezi MCC je mutace v genu pro virový T antigen a integrace viru do hostitelského genomu, které jsou neslučitelné s produktivním virovým cyklem. Protože neexistují žádné zvířecí modely MCC, používají se pro studium životního cyklu a karcinogeneze MCPyV buněčné in vitro modely využívající především fibroblasty a 293 buňky, a 293T buňky, které jsou odvozené z embryonálních buněk ledvin. Cílem této diplomové práce je ustavit v našich laboratorních podmínkách in vitro model pro studium MCPyV pomocí transfekce fibroblastů, 293T a 293 buněk virovými izoláty získanými z kůže zdravých jedinců, a studovat replikaci virového genomu a expresi virových mRNA, popř. sledovat produkci virionů, pomocí metod qPCR, RT-qPCR a elektronového mikroskopu.

Merkel cell carcinoma (MCC) is a rare aggressive skin neuroendocrine tumor with metastasis. More than 80% of MCCs are associated with infection of Merkel cell polyomavirus (MCPyV). A major step in the carcinogenesis of MCC is the mutation in the T antigen gene and the integration of the virus into the host genome, which are incompatible with the productive viral cycle. Since there are no animal models of MCC, in vitro models using fibroblasts, 293 cells and 293T cells which are derived from embryonic kidney cells are used to study the life cycle and carcinogenesis of MCPyV. The aim of this thesis is to establish in our laboratory an in vitro model for the study of MCPyV by transfection fibroblasts and/or 293 and/or 293T cells with virus isolates obtained from the skin of healthy individuals and monitor viral gene expresion, replication and virus production using qPCR, RT-qPCR, and electron microscopy.
