Diplomový projekt je součástí širšího výzkumu laboratoře, který se zabývá možným využitím virových částic (VLPs) odvozených od myšího polyomaviru jako transportních systémů pro dopravu terapeutik do buněk. Pro dosažení optimálního účinku je nutné zamezit hromadění virových nosičů v lysozomech a zajistit řízené uvolnění terapeutika uvnitř buněk. Peptidy bohaté na histidin představují zajímavou možnost, jak zvýšit únik nosičů do cytosolu. Tato práce bude mít za hlavní cíl zjistit, jak polyhisitdinové peptidy ovlivňují účinnost transdukce heterologní DNA ve virových částicích modifikovaných polyhistidinem, zda usnadňují jejich únik do cytosolu a porovnat jejich účinek s ostatními membránově aktivními peptidy studovanými v laboratoři (LAH4, R8).
Preliminary scope of work in English
The diploma project is part of a wider research of the laboratory, which deals with the possible use of viral particles (VLPs) derived from murine polyomavirus as transport systems for the delivery of therapeutics into cells. To achieve the optimal effect, it is necessary to prevent the accumulation of viral carriers in lysosomes and to ensure controlled release of the therapeutic within the cells.
