Viry vstupují do hostitelských buněk pomocí endocytózy. Nízké pH v endosomech iniciuje konformační změny ve strukturních proteinech virionů. Tyto změny, které destabilizují kapsidy nebo vedou k fůzi virových a endosomálních membrán jsou obvykle nutné pro účinné zahájení infekce. Látky, které snižují vnitrobuněčné pH také snižují infektivitu virů, u kterých je infekce na pH závislá. Naopak, mírně kyselé extracelulární prostředí při infekci zvyšuje infektivitu mnoha virů. Tato bakalářská práce by měla přehledně shrnout publikovaná data ukazující důležitost pH při infekci u různých virových čeledí a popsat molekulární mechanismy, které mohou vysvětlit efekty, které má nízké pH na virovou infekci. |
Preliminary scope of work in English |
Viruses enter host cells by endocytosis. The low pH of endosomes triggers conformational changes in structural proteins of virions. These changes that destabilize capsids or mediate fusion of the viral and endosomal membranes are usually required for successful initiation of infection. Drugs that increase intracellular pH have been shown to decrease infectivity of pH-dependent viruses. On contrary, the mildly acidic environment during initiation of infection increases infectivity of many viruses. This bachelor thesis should focus on the publised data for different viral families that describe the relevance of pH balance during viral infection and describe the putative molecular mechanism(s) that underlay low-pH effect on viral infection. |
