Preliminary scope of work
Usměrněná evoluce je účinný přístup, který dovoluje relativně rychlou selekci a izolaci virových variant s novými a terapeuticky cennými vlastnostmi. Cílem této diplomové práce je vytvořit kvalitní a vysoce diversifikovanou knihovnu polyomavirových variant mutovaných v hlavním kapsidovém proteinuVP1. Pro přípravu mutant v Cre/loxP systému vyvinutém v laboratoři budou použity metody “error-prone PCR” a “DNA shuffling”.
Preliminary scope of work in English
Directed evolution is a powerful approach that enables relatively rapid selection and isolation of virus variants with novel and therapeutically valuable properties. The main goal of this diploma thesis is to create the high quality, high diversity libraries of polyomavirus variants mutated in main capsid protein VP1. The error-prone PCR and DNA shuffling methods will be used for preparation of mutants in Cre/loxP system developed in the laboratory.
