Lidský polyomavirus MCPyV byl objeven v roce 2008 ve vzácném kožním nádoru karcinomu z Merkelových buněk. MCPyV DNA je přítomna v 80% karcinomu z Merkelových buněk. Dalšími rizikovými faktory vzniku nádoru jsou, kromě věku, expozice UV, pohlaví, rasa a imunosuprese. Virus je velmi často přítomen také u zdravých jedinců. Sérologické studie ukazují, že k infekci dochází již raném v dětství, a v dospělé populaci je MCPyV běžný. Nicméně, zatím existuje málo informací o přítomnosti MCPyV v krvi imunosuprimovaných jedinců. Cílem této studie je zjistit přítomnost MCPyV DNA u pacientů po transplantaci kmenových krvetvorných buněk (HSCT) a u zdravých jedinců. Druhým cílem této studie je určit sekvence virového genu pro T antigen u zdravých jedinců a pacientů po HSCT a porovnat tyto sekvence s T antigenem z karcinomů z Merkelových buněk, kde se velmi často nachází delece nebo mutace tohoto genu
Preliminary scope of work in English
In the year 2008, the Merkel cell polyomavirus (MCPyV), associated with Merkel cell carcinoma (MCC), rare but very aggressive neuroendocrine cancer, was identified. MCPyV DNA is being found in about 80% of MCC. Risk factors for MCC are older age, immunosuppression and skin exposure to UV light. MCPyV DNA is also found in healthy individuals. Seroprevalence data suggest that MCPyV infection occurs during early childhood and that this virus is ubiquitous among adults. However, there is little information about the presence of MCPyV in the blood of immunocompromised individuals. The aim of this study is to find the prevalence of MCPyV DNA in patients after hematopoietic stem cell transplantation and in healthy individuals. The second objective of this study is to determine the sequence of the viral gene for the T antigen in healthy subjects and patients after HSCT and compare these sequences with the T antigen of Merkel cell carcinoma, which often share deletion or mutation of this gene.
