Preliminary scope of work
Porothramycin je antibiotikum anthramycinové skupiny a je produkován kmenem Streptomyces albus. Předpokládá se, že některé geny účastnící se syntézy porothramycinu lze využít k?přípravě nových antibiotik metodou kombinatoriální biosyntézy. Základním předpokladem k?tomu je znalost biosyntetické dráhy této látky. Za účelem jejího studia je nezbytné vyvinout metodu chemické analýzy tohoto antibiotika. Nejprve bude na základě literárání rešerše nalezeno médium vhodné jak pro růst bakterie, tak pro produkci porothtramycinu. Dále bude vyvinuta metoda izolace porothramycinu z?kultivačního média a následně bude vyvinuta metoda stanovení tohoto analytu pomocí ultraúčinné kapalinové chromatografie s?UV a hmotnostní detekcí. Poté bude metoda aplikována na reálné vzorky.
Preliminary scope of work in English
Porothramycin is the antibiotic of anthramycin group and is produced by Streptomyces albus. It is considered that some genes involved in the synthesis of porothramycin can be used for the preparation of novel antibiotics by means of combinatorial biosynthesis. Precise knowledge of the porothramycoin biosynthesic pathway represents the main prerequisite for the approach of combinatorial biosynthesis. For this purpose, it is necessary to develop methods of chemical analysis of this antibiotic. Firstly, cultivation broth suitable for both bacterial growth and porothtramycin production will be found. Then, method for the analyte isolation from cultivation broth and subsequently the method for porothramycin analysis by ultra-performance liquid chromatography with UV and mass spectroscopy detection will be developed. Lastly, the method will be applied for real samples.
